Hepatoprotective effects of l-methionine and methanolic extract of Silybum marianum against nickel induced liver injury and oxidative stress in rats

Abstract
Background: Nickel is definitely regarded as a potential real human toxicant, having the ability to form carcinomas. Therefore, there exists a constant demand for exogenous antioxidants to be able to avoid this oxidative stress. Aims and Objectives: To investigate the hepatoprotective effect of methanolic extract of Silybummarianum (Sil) and L-methionine against nickel-induced oxidative damage in experimental rats. Methods: Male albinos Wistar rats divided into six groups of seven each, the first was served as a control, the remaining groups, respectively treated with: silymarin, methionine (100 mg/kgbody weight by gavage), nickel sulfate (20 mg/kg body weight by i.p) and a combination of nickel plus silymarin (Ni+Sil) and nickel plus methionine (Ni+Met). The experiment lasted 21 days. Results: Obtained results revealed that administration of nickel caused a significant increase of ASAT, LDH and total bilirubin (p<0.001), ALAT (p<0.01) and ALP (p<0.05) compared to the control group. Furthermore, the exposure to nickel significantly increased MDA level (p<0.001), decreased GSH concentration and hepatic GPx, CAT and SOD activities. However, the treatment with Silymarin or L-Met reversed biochemical indices of liver injury and enhanced the antioxidant status. Silymarin and L-methionine also conferred variable degrees of tissue protection. Conclusions: The results showed that either the methanolic extract of milk thistle or L-methionine possessedhepatoprotective activity against nickel-induced oxidative liver injury in rats.
